03. 1P2C Assembly Guide

Welcome to the 1P2C Microscope assembly guide.

All parts for assembly can be found on page one of the 1P2C Parts List.

General Assembly Tips & Tricks

  • The filters are directional. A good rule of thumb to ensure that the filters are placed in the correct orientation is that the reflective side faces the BLUE LED. Typically the filters will have arrows indicating directionality but over time they may wear off.
  • Do not use metal tweezers/instruments on the filters or lenses as this may damage them. Carbon-tipped tweezers work the best in our experience.
  • The blunt wooden end of cotton swabs work well to push the filters and lenses in straight after positioning with tweezers.

Filters


01-EM 02-BS

Insert the emission filter and the emission dichroic with the reflective sides facing down.



03-EX 04-LXPR

Insert the excitation filter with the reflective side facing the LED slots and
insert the excitation dichroic with the reflective side facing upward.



Lenses


05-Ach2

Insert the achromat with the convex side down, taking care to ensure that it is level.



06-drum

Insert both drum lenses into the holes in the LED slots. The drum lenses are bidirectional.



07-GRIN

Insert the GRIN objective until the top of the lens is flush with the inside floor of the miniscope.



Electronics


08-CMOS 09-LED1

Screw on the CMOS Mount and secure with set screw, then slide the LEDs into their slots.

To focus the CMOS:

Screw it down to the bottom or second to bottom thread then place it above a test slide that is illuminated from underneath. Slightly adjust the height of the CMOS until the smallest target on the slide is in focus and tighten the set screw. Note that small adjustments may be necessary to refocus on your target cells in vivo.
