AFAIK, you can use command line to do it: python manage.py assets build

If you want to do it @before_first_request, you can call it from Python code: manager.handle('assets', ('build,)) # untested

Current Flask-Assets==0.8 has a big issue with Flask-Script>=0.6 (miracle2k/flask-assets#52), but looks like it is fixed on master branch.
