Essentially the disulfide the Cys to be mutagenised needs to be "turned off" from detection, while we do want to keep any other disulfides in the protein. Unclear what/if anything needs to happen to the other side of the disulfide bridge. Anders I think you had a good plan for this already, I'm just putting it here to make sure it doesn't get lost.
